Biomechanical and biochemical assessment of YB-1 expression in A375 melanoma cell line: Exploratory study

Malignant melanoma is the most lethal form of skin cancer. Y-box binding protein 1 (YB-1) plays a prominent role in mediating metastatic behavior by promoting epithelial-to-mesenchymal transition (EMT). Migratory cells exhibit two major migration modes: elongated mesenchymal or rounded amoeboid. Using A375 cell line and YB-1 knock-out model, we aimed to elucidate biochemical biomechanical changes signaling pathways context metastases. We subjected parental atomic force microscopy (stiffness determination), immunolabelling, proteome analysis. found that expressing were significantly stiffer compared corresponding line. Our study demonstrated constitutive expression appears be closely related known biomarkers transition, nestin, vimentin, resulting phenotype, as well wide array proteins involved RNA, ribosomes, spliceosomes. resulted nestin depletion lower vimentin expression, global upregulation cytoskeleton migration. both morphological features drivers mesenchymal/ameboid Melanoma highly plastic, adaptable, aggressive tumor entity, capable exhibiting characteristics different migratory modes.
